Anna Maria College was born as the result of a woman named Esther Blondin who sought to care for and educate the people of Quebec, Canada during the mid 19th century. Through Blondin's efforts, the congregation of the Sisters of Saint Anne was developed with the purpose of teaching the underprivileged about religion, reading, writing and arithmetic. As a result, Blondin is now known as Mother Marie Anne. In her honor, the Sisters of Saint Anne founded Anna Maria College in 1946 to continue her educational mission. Initially located in Marlborough, Massachusetts, the school shifted to Paxton in 1952.
According to its stated mission, "Anna Maria College, a Catholic institution of higher learning, recognizes its obligation to serve its immediate community, the Commonwealth of Massachusetts, the nation and the world through the provision of education, the preservation of learning and the sponsorship of research."
